Bink 是目前 Windows 游戏中应用最广泛的动画格式(简写为 .bik格式),Bink Video文件以.bik为后缀名,被广泛的运用于游戏领域,具体表现为开场动画、任务剧情动画以及版权声明动画等等。

而战地2的游戏背景视频就是采用的bik格式,那么如果你在创作模组的时候,想加入一个自己制作的动画视频,那么Rad Video Tools就派上了用场。所谓工欲善其事,必先利其器。一个优秀的软件可以让你的工作事半功倍,而Rad Video Tools就是一款在bik视频转码领域非常出色的软件之一。

而关于这个软件的使用教程网络是少之又少,本文就分享作者关于这个软件的使用的一些经验。

使用前的准备:

1,下载并安装Rad Video Tools(即本文所讲的这个软件,下载地址):

2,下载并安装Quiktime (下载地址)

注意:由于Rad video tools是一个古老的软件工具,而它设计之初的转换功能依赖于苹果公司旗下的这个Quiktime播放器,必须安装。

* 在英文原版中,如果没有安装Quiktime播放器时进行的bik格式转换将弹出上图中的错误提示。而在中文汉化版中则直接跳过了这个错误,提示打开文件错误。

使用方法&转换教程:

1,打开RAD Video Tools,并选择想要转换的视频文件,并点击转换按钮。如下图:

* 注意视频文件不能使用中文命名,否则会出现打开文件错误;

2,之后会跳到信息的调整界面,我们如果不需要特别修改,就直接点击转换即可,如下图:

3,之后就是漫长的转换过程,耐心等待完毕后,会在原视频文件文件夹自动产生一个 .bik的视频文件

4,最终完成。

然后我们看看游戏里的效果:

这样一个bik的视频就制作完成了。需要说明的是,如果你需要调整一些视频的高级细节、例如码率等,推荐使用其他软件转换任意mp4或者avi后,在通过这个软件转换为bik。如果需要直接播放这个视频,推荐使用VLC播放器、KMplayer、或迅雷播放器都可以。

为毛转换得bik视频播放速度比原来的快很多啊,像2倍速度在播放一样

关注采用bik文件格式的游戏很多,如大宇双剑系列、热血无赖等,体积较小,但资源占用较高。bik(Bink Video)是一种媒体文件格式,一种具有高压缩比、高画面质量、高音频质量,更优于MPEG2编码格式的优秀视频文件格式。Bink Video文件以bik为后缀名,被广泛的运用于计算机电子游戏(Computer Electronic Game)上,具体表现为开场动画、任务剧情动画以及版权声明动画等等。一般人会觉得很少有媒体播放工具支持Bink Video格式的文件,因为缺乏解码系统。游戏中播放Bink Video通常会使用游戏自带的解码系统,或是自行研发,或是采用官方发布版本。迅雷看看、暴风影音、QQ影音、射手影音、BaNaNa Binkplay、RAD Video Tools以及KMPlayer都可以可以播放bik文件.(注:其中KM要经过设置才能打开的 在参数设置中的关联选项中添加BIK扩展名,再点应用到系统即可,如果偏爱其他媒体播放器的话,试试这个方法应该可以)至于压制转码很好压,用小丸就能压

AbstractBIK is the founding member of the BH3-only family pro-apoptotic proteins. BIK is predominantly localized in the ER and induces apoptosis through the mitochondrial pathway by mobilizing calcium from the ER to the mitochondria and remodeling the mitochondrial cristae. BIK-mediated apoptosis is mediated by selective activation of BAX. BIK also induces non-apoptotic cell death in certain cell types by unknown mechanisms. BIK is non-essential for animal development, but appears to be functionally redundant for certain developmental functions with BIM. BIK is implicated in the selection of mature B cells in humans. BIK is a pro-apoptotic tumor suppressor in several human tissues and its expression in cancers is prevented by chromosomal deletions encompassing the Bik locus or by epigenetic silencing. BIK appears to be a critical effector in apoptosis induced by toxins, cytokines and virus infection. Several anti-cancer drugs transcriptionally activate Bik gene expression through transcriptional pathways dependent on factors such as E2F and p53 or by removal of epigenetic marks on the chromatin. BIK appears to be a prominent target for anti-cancer drugs that inhibit proteasomal functions. BIK has also been used as a therapeutic molecule in gene therapy-based approaches to treat difficult cancers.

Western Blotting Protocol

For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

NOTE: Please refer to primary antibody product webpage for recommended antibody dilution.

A. Solutions and Reagents

From sample preparation to detection, the reagents you need for your Western Blot are now in one convenient kit: #12957 Western Blotting Application Solutions Kit

NOTE: Prepare solutions with reverse osmosis deionized (RODI) or equivalent grade water.

20X Phosphate Buffered Saline (PBS): (#9808) To prepare 1 L 1X PBS: add 50 ml 20X PBS to 950 ml dH2O, mix.

10X Tris Buffered Saline (TBS): (#12498) To prepare 1 L 1X TBS: add 100 ml 10X to 900 ml dH2O, mix.

1X SDS Sample Buffer: Blue Loading Pack (#7722) or Red Loading Pack (#7723) Prepare fresh 3X reducing loading buffer by adding 1/10 volume 30X DTT to 1 volume of 3X SDS loading buffer. Dilute to 1X with dH2O.

10X Tris-Glycine SDS Running Buffer: (#4050) To prepare 1 L 1X running buffer: add 100 ml 10X running buffer to 900 ml dH2O, mix.

10X Tris-Glycine Transfer Buffer: (#12539) To prepare 1 L 1X Transfer Buffer: add 100 ml 10X Transfer Buffer to 200 ml methanol + 700 ml dH2O, mix.

10X Tris Buffered Saline with Tween® 20 (TBST): (#9997) To prepare 1 L 1X TBST: add 100 ml 10X TBST to 900 ml dH2O, mix.

Nonfat Dry Milk: (#9999).

Blocking Buffer: 1X TBST with 5% w/v nonfat dry milk; for 150 ml, add 7.5 g nonfat dry milk to 150 ml 1X TBST and mix well.

Wash Buffer: (#9997) 1X TBST.

Bovine Serum Albumin (BSA): (#9998).

Primary Antibody Dilution Buffer: 1X TBST with 5% BSA; for 20 ml, add 1.0 g BSA to 20 ml 1X TBST and mix well.

Biotinylated Protein Ladder Detection Pack: (#7727).

Blue Prestained Protein Marker, Broad Range (11-250 kDa): (#59329).

Blotting Membrane and Paper: (#12369) This protocol has been optimized for nitrocellulose membranes. Pore size 0.2 µm is generally recommended.

Secondary Antibody Conjugated to HRP: Anti-rabbit IgG, HRP-linked Antibody (#7074).

Detection Reagent: SignalFire™ ECL Reagent (#6883).

B. Protein Blotting

A general protocol for sample preparation.

Treat cells by adding fresh media containing regulator for desired time.

Aspirate media from cultures; wash cells with 1X PBS; aspirate.

Lyse cells by adding 1X SDS sample buffer (100 µl per well of 6-well plate or 500 µl for a 10 cm diameter plate). Immediately scrape the cells off the plate and transfer the extract to a microcentrifuge tube. Keep on ice.

Sonicate for 10–15 sec to complete cell lysis and shear DNA (to reduce sample viscosity).

Heat a 20 µl sample to 95–100°C for 5 min; cool on ice.

Microcentrifuge for 5 min.

Load 20 µl onto SDS-PAGE gel (10 cm x 10 cm).

NOTE: Loading of prestained molecular weight markers (#59329, 10 µl/lane) to verify electrotransfer and biotinylated protein ladder (#7727, 10 µl/lane) to determine molecular weights are recommended.

Electrotransfer to nitrocellulose membrane (#12369).

C. Membrane Blocking and Antibody Incubations

NOTE: Volumes are for 10 cm x 10 cm (100 cm2) of membrane; for different sized membranes, adjust volumes accordingly.

I. Membrane Blocking

(Optional) After transfer, wash nitrocellulose membrane with 25 ml TBS for 5 min at room temperature.

Incubate membrane in 25 ml of blocking buffer for 1 hr at room temperature.

Wash three times for 5 min each with 15 ml of TBST.

II. Primary Antibody Incubation

Incubate membrane and primary antibody (at the appropriate dilution and diluent as recommended in the product webpage) in 10 ml primary antibody dilution buffer with gentle agitation overnight at 4°C.

Wash three times for 5 min each with 15 ml of TBST.

Incubate membrane with Anti-rabbit IgG, HRP-linked Antibody (#7074 at 1:2000) and anti-biotin, HRP-linked Antibody (#7075 at 1:1000–1:3000) to detect biotinylated protein markers in 10 ml of blocking buffer with gentle agitation for 1 hr at room temperature.

Wash three times for 5 min each with 15 ml of TBST.

Proceed with detection (Section D).

D. Detection of Proteins

Directions for Use:

Wash membrane-bound HRP (antibody conjugate) three times for 5 minutes in TBST.

Prepare 1X SignalFire™ ECL Reagent (#6883) by diluting one part 2X Reagent A and one part 2X Reagent B (e.g. for 10 ml, add 5 ml Reagent A and 5 ml Reagent B). Mix well.

Incubate substrate with membrane for 1 minute, remove excess solution (membrane remains wet), wrap in plastic and expose to X-ray film.

* Avoid repeated exposure to skin.

Bik Antibody detects endogenous levels of Bik protein. It does not cross-react with other Bcl-2 family members.

Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues in the amino-terminus of Bik. Antibodies are purified by protein A and peptide affinity chromatography.

Background

Bik/Nbk (Bcl-2-interacting killer/natural born killer) is a potent pro-apoptotic protein belonging to a group of Bcl-2 family members that includes Bad, Bid, Bim, Hrk, and Noxa, containing a BH3 domain but lacking other conserved domains, BH1 or BH2 (1,2). Functionally, Bik is able to bind to and antagonize anti-apoptotic Bcl-2 family members including Bcl-2, Bcl-xL, and viral homologs E1B-19K and EBV-BHFR1. The BH3 domain of Bik is essential for its apoptotic activity and interaction with survival proteins (3). Phosphorylation of Bik is correlated with an increase in its pro-apoptotic activity (4).
